NM_024675.4(PALB2):c.389A>C (p.His130Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H130P variant (also known as c.389A>C), located in coding exon 4 of the PALB2 gene, results from an A to C substitution at nucleotide position 389. The histidine at codon 130 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
