Uncertain significance for Osteogenesis imperfecta type 7 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006371.5(CRTAP):c.170G>A (p.Gly57Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with aspartic acid at codon 57 of the CRTAP protein (p.Gly57Asp). The glycine residue is moderately conserved and there is a moderate physicochemical difference between glycine and aspartic acid. This variant is present in population databases (rs747689884, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with CRTAP-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
