Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001244008.2(KIF1A):c.3829C>T (p.Arg1277Trp), citing Ambry Variant Classification Scheme 2023: The p.R1277W variant (also known as c.3829C>T), located in coding exon 36 of the KIF1A gene, results from a C to T substitution at nucleotide position 3829. The arginine at codon 1277 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
