This variant lies in the AP4B1 gene (transcript NM_001253852.3) at coding-DNA position 1258, where T is replaced by C; at the protein level this means replaces cysteine at residue 420 with arginine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
