NM_177438.3(DICER1):c.2509A>G (p.Met837Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M837V variant (also known as c.2509A>G), located in coding exon 15 of the DICER1 gene, results from an A to G substitution at nucleotide position 2509. The methionine at codon 837 is replaced by valine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
