NM_006361.6(HOXB13):c.599C>T (p.Ala200Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A200V variant (also known as c.599C>T), located in coding exon 1 of the HOXB13 gene, results from a C to T substitution at nucleotide position 599. The alanine at codon 200 is replaced by valine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
