Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_014000.3(VCL):c.1195A>G (p.Asn399Asp), citing Ambry Variant Classification Scheme 2023: The p.N399D variant (also known as c.1195A>G), located in coding exon 10 of the VCL gene, results from an A to G substitution at nucleotide position 1195. The asparagine at codon 399 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
