This variant lies in the PSTPIP1 gene (transcript NM_003978.5) at coding-DNA position 469, where G is replaced by T; at the protein level this means replaces alanine at residue 157 with serine — a missense variant. Submitter rationale: This sequence change replaces alanine with serine at codon 157 of the PSTPIP1 protein (p.Ala157Ser). The alanine residue is weakly conserved and there is a moderate physicochemical difference between alanine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with PSTPIP1-related conditions.
