It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SPATA5 are known to be pathogenic (PMID: 26299366). This variant is present in population databases (rs773729755, gnomAD 0.003%). This premature translational stop signal has been observed in individual(s) with epilepsy and developmental delay (PMID: 29343804). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 844710). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
