Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000363.5(TNNI3):c.322G>A (p.Asp108Asn), citing Ambry Variant Classification Scheme 2023: The p.D108N variant (also known as c.322G>A), located in coding exon 6 of the TNNI3 gene, results from a G to A substitution at nucleotide position 322. The aspartic acid at codon 108 is replaced by asparagine, an amino acid with highly similar properties. This alteration has been reported in a cohort of subjects with cardiomyopathy (Smith E et al. J Am Heart Assoc, 2022 May;11:e024501). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
