NM_001369.3(DNAH5):c.10658G>A (p.Arg3553Gln) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R3553Q variant (also known as c.10658G>A), located in coding exon 63 of the DNAH5 gene, results from a G to A substitution at nucleotide position 10658. The arginine at codon 3553 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
