NM_004656.4(BAP1):c.451C>T (p.His151Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H151Y variant (also known as c.451C>T), located in coding exon 7 of the BAP1 gene, results from a C to T substitution at nucleotide position 451. The histidine at codon 151 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
