Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004385.5(VCAN):c.157C>T (p.Pro53Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 53 of the VCAN protein (p.Pro53Ser). This variant is present in population databases (rs201466502, gnomAD 0.02%). This missense change has been observed in individuals with clinical features of retinitis pigmentosa (internal data). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 843872). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign".
