Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_194277.3(FRMD7):c.998A>T (p.His333Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine with leucine at codon 333 of the FRMD7 protein (p.His333Leu). The histidine residue is weakly conserved and there is a moderate physicochemical difference between histidine and leucine. This variant is present in population databases (rs781757793, ExAC 0.03%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals with FRMD7-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
