NM_177438.3(DICER1):c.753_755dup (p.Cys251_Glu252insAsp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the DICER1 gene (transcript NM_177438.3) at coding-DNA position 753 through coding-DNA position 755, duplicating 3 bases. Submitter rationale: The c.753_755dupTGA variant (also known as p.C251_E252insD) is located in coding exon 6 of the DICER1 gene. This variant results from an in-frame duplication of 3 nucleotides at positions 753 to 755. This results in the insertion of an aspartic acid residue between codons 251 and 252. This amino acid region is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
