Uncertain significance for Susceptibility to respiratory infections associated with CD8alpha chain mutation — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001768.7(CD8A):c.279G>T (p.Arg93Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with serine, which is neutral and polar, at codon 93 of the CD8A protein (p.Arg93Ser). This variant is present in population databases (rs2229240, gnomAD 0.2%). This variant has not been reported in the literature in individuals affected with CD8A-related conditions. ClinVar contains an entry for this variant (Variation ID: 843539). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
