Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.29A>T (p.Glu10Val), citing Ambry Variant Classification Scheme 2023: The p.E10V variant (also known as c.29A>T), located in coding exon 2 of the PMS2 gene, results from an A to T substitution at nucleotide position 29. The glutamic acid at codon 10 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
