NM_177438.3(DICER1):c.5464G>A (p.Asp1822Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1822N variant (also known as c.5464G>A), located in coding exon 24 of the DICER1 gene, results from a G to A substitution at nucleotide position 5464. The aspartic acid at codon 1822 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
