This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 1739, where A is replaced by G; at the protein level this means replaces asparagine at residue 580 with serine — a missense variant. Submitter rationale: Variant summary: RYR1 c.1739A>G (p.Asn580Ser) results in a conservative amino acid change located in the RIH domain (IPR000699) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 251462 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1739A>G in individuals affected with Malignant Hyperthermia Susceptibility or other RYR1-related disorders and no experimental evidence demonstrating its impact on protein function have been reported.
