Uncertain significance for Distal hereditary motor neuropathy type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_205836.3(FBXO38):c.1970T>A (p.Met657Lys), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This sequence change replaces methionine with lysine at codon 657 of the FBXO38 protein (p.Met657Lys). The methionine residue is moderately conserved and there is a moderate physicochemical difference between methionine and lysine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with FBXO38-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
