Uncertain significance for Peroxisome biogenesis disorder, complementation group K — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004565.3(PEX14):c.199T>C (p.Phe67Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine with leucine at codon 67 of the PEX14 protein (p.Phe67Leu). The phenylalanine residue is moderately conserved and there is a small physicochemical difference between phenylalanine and leucine. This variant is present in population databases (rs751540499, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with PEX14-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
