Submitter rationale: The p.R143Q variant (also known as c.428G>A), located in coding exon 4 of the RAF1 gene, results from a G to A substitution at nucleotide position 428. The arginine at codon 143 is replaced by glutamine, an amino acid with highly similar properties, and is located in the cysteine-rich domain. An indirect in vitro functional assay suggested this variant may result in a slight activation of RAF1 enzymatic activity, but the physiological relevance of the result is unclear (Winkler DG et al.
