Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004360.5(CDH1):c.2263T>C (p.Tyr755His), citing Ambry Variant Classification Scheme 2023: The p.Y755H variant (also known as c.2263T>C), located in coding exon 14 of the CDH1 gene, results from a T to C substitution at nucleotide position 2263. The tyrosine at codon 755 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
