NM_002335.4(LRP5):c.250G>A (p.Glu84Lys) was classified as Uncertain significance for Exudative vitreoretinopathy 4 by Johns Hopkins Genomics, Johns Hopkins University, citing ACMG Guidelines, 2015: This LRP5 variant (rs201119420) is rare (<0.1%) in a large population dataset (gnomAD: 4/251310 total alleles; 0.0016%; no homozygotes) and has been reported in ClinVar. Of two bioinformatics tools queried, one predicts that the substitution would be possibly damaging, while another predicts that it would be tolerated. The glutamic acid residue at this position is strongly conserved across the vertebrate species assessed. Bioinformatic analysis predicts that this missense variant would not affect normal exon 2 splicing, although this has not been confirmed experimentally to our knowledge. Due to insufficient evidence that this variant is deleterious, we consider the clinical significance of c.250G>A (p.Glu84Lys) to be uncertain at this time.
