NM_000238.4(KCNH2):c.2853C>A (p.Ser951Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S951R variant (also known as c.2853C>A), located in coding exon 12 of the KCNH2 gene, results from a C to A substitution at nucleotide position 2853. The serine at codon 951 is replaced by arginine, an amino acid with dissimilar properties, and is located in the C-terminal region of the protein. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
