Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001365999.1(SZT2):c.5686C>G (p.Pro1896Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline with alanine at codon 1839 of the SZT2 protein (p.Pro1839Ala). The proline residue is highly conserved and there is a small physicochemical difference between proline and alanine. This variant is present in population databases (rs758455492, ExAC 0.003%). This variant has not been reported in the literature in individuals affected with SZT2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C25".
