NM_004656.4(BAP1):c.1895T>C (p.Leu632Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L632P variant (also known as c.1895T>C), located in coding exon 15 of the BAP1 gene, results from a T to C substitution at nucleotide position 1895. The leucine at codon 632 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
