Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000071.3(CBS):c.1325G>A (p.Gly442Asp), citing Ambry Variant Classification Scheme 2023: The p.G442D variant (also known as c.1325G>A), located in coding exon 12 of the CBS gene, results from a G to A substitution at nucleotide position 1325. The glycine at codon 442 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
