Uncertain significance for Trichorhinophalangeal syndrome, type III; Trichorhinophalangeal dysplasia type I — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014112.5(TRPS1):c.275T>C (p.Val92Ala), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TRPS1 gene (transcript NM_014112.5) at coding-DNA position 275, where T is replaced by C; at the protein level this means replaces valine at residue 92 with alanine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with TRPS1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces valine with alanine at codon 92 of the TRPS1 protein (p.Val92Ala). The valine residue is weakly conserved and there is a small physicochemical difference between valine and alanine.
