NM_000465.4(BARD1):c.1903+2dup was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1903+2dupT intronic variant is located two nucleotides after coding exon 9 in the BARD1 gene. This variant results from a duplication of one nucleotide at nucleotide position 1903. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site; however, direct evidence is insufficient at this time (Ambry internal data). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
