Uncertain significance for Distal hereditary motor neuropathy type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_205836.3(FBXO38):c.617G>C (p.Cys206Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine, which is neutral and slightly polar, with serine, which is neutral and polar, at codon 206 of the FBXO38 protein (p.Cys206Ser). This variant is present in population databases (rs757064226, gnomAD 0.0009%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant disrupts the p.Cys206 amino acid residue in FBXO38. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 24207122). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt FBXO38 protein function. ClinVar contains an entry for this variant (Variation ID: 839326). This variant has not been reported in the literature in individuals affected with FBXO38-related conditions.
