NM_001114753.3(ENG):c.293T>A (p.Val98Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V98D variant (also known as c.293T>A), located in coding exon 3 of the ENG gene, results from a T to A substitution at nucleotide position 293. The valine at codon 98 is replaced by aspartic acid, an amino acid with highly dissimilar properties. In our internal cohort, this variant was identified in an individual with epistaxis, telangiectasias, and hepatic arteriovenous malformations; it segregated with disease in two relatives of the proband. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on available evidence to date, the clinical significance of this alteration remains unclear.
