NM_000548.5(TSC2):c.567T>A (p.Cys189Ter) was classified as Pathogenic for Tuberous sclerosis 2 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant has been observed in individuals with diagnosis or suspicion of tuberous sclerosis complex (PMID: 25782670, Invitae), in at least one of whom it has been observed de novo. This sequence change creates a premature translational stop signal (p.Cys189*) in the TSC2 gene. It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). Loss-of-function variants in TSC2 are known to be pathogenic (PMID: 10205261, 17304050). For these reasons, this variant has been classified as Pathogenic.
