Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000251.3(MSH2):c.968_971dup (p.Gln324fs), citing Ambry Variant Classification Scheme 2023: The c.968_971dupCTCA pathogenic mutation, located in coding exon 6 of the MSH2 gene, results from a duplication of CTCA at nucleotide position 968, causing a translational frameshift with a predicted alternate stop codon (p.Q324Hfs*10). This alteration was identified in an individual diagnosed with colorectal cancer (Chubb D et al. J Clin Oncol, 2015 Feb;33:426-32). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
