Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 4e-06 in 251492 control chromosomes (gnomAD). c.2947A>G has been observed in multiple individuals affected with Stargardt Disease (Ernest_2009, Bax_2019, Lambertus_2014). These data indicate that the variant is very likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function and this variant affected ABCA4 protein function (Molday_2018).
