This variant lies in the DST gene (transcript NM_001723.7) at coding-DNA position 3373, where C is replaced by T; at the protein level this means replaces arginine at residue 1125 with cysteine — a missense variant. Submitter rationale: This variant is present in population databases (rs776487965, ExAC 0.01%). This variant has not been reported in the literature in individuals with DST-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
