This variant has not been reported in the literature in individuals with DNAAF5-related conditions. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This sequence change replaces threonine with isoleucine at codon 533 of the DNAAF5 protein (p.Thr533Ile). The threonine residue is weakly conserved and there is a moderate physicochemical difference between threonine and isoleucine.
