Uncertain Significance for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma — the classification assigned by All of Us Research Program, National Institutes of Health to NM_004415.4(DSP):c.7685_7687del (p.Ser2562del), citing ACMG Guidelines, 2015. This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 7685 through coding-DNA position 7687, deleting 3 bases; at the protein level this means deletes serine at residue 2562. Submitter rationale: This variant causes a deletion of one amino acid at codon 2562 of the DSP protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has been identified in 4/251492 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
