Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012193.4(FZD4):c.1487G>A (p.Trp496Ter), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). For these reasons, this variant has been classified as Pathogenic. This variant disrupts the C-terminus of the FZD4 protein. Other variant(s) that disrupt this region (p.Thr500fs*12, p.Gln505*, p.Leu501fs*32) have been observed in individuals with FZD4-related conditions (PMID: 15223780). This suggests that this may be a clinically significant region of the protein. ClinVar contains an entry for this variant (Variation ID: 837460). This premature translational stop signal has been observed in individuals with familial exudative vitreoretinopathy (PMID: 19324841, 30452590). It has also been observed to segregate with disease in related individuals. This sequence change creates a premature translational stop signal (p.Trp496*) in the FZD4 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 42 amino acid(s) of the FZD4 protein.
