NM_005585.5(SMAD6):c.106G>T (p.Asp36Tyr) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D36Y variant (also known as c.106G>T), located in coding exon 1 of the SMAD6 gene, results from a G to T substitution at nucleotide position 106. The aspartic acid at codon 36 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
