Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.5324del (p.Phe1774_Leu1775insTer), citing Ambry Variant Classification Scheme 2023: The c.5324delT pathogenic mutation, located in coding exon 35 of the ATM gene, results from a deletion of one nucleotide at nucleotide position 5324, causing a translational frameshift with a predicted alternate stop codon (p.L1775*). This variant has been observed in individual(s) with ataxia-telangiectasia (Li A et al. Am J Med Genet, 2000 May;92:170-7). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
