Uncertain significance for Dyskeratosis congenita — the classification assigned by Ambry Genetics to NM_198253.3(TERT):c.2411G>A (p.Gly804Asp), citing Ambry Variant Classification Scheme 2023: The p.G804D variant (also known as c.2411G>A), located in coding exon 8 of the TERT gene, results from a G to A substitution at nucleotide position 2411. The glycine at codon 804 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
