NM_005359.6(SMAD4):c.533C>A (p.Ser178Ter) was classified as Pathogenic for Juvenile polyposis syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ser178*) in the SMAD4 gene. It is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in SMAD4 are known to be pathogenic (PMID: 16152648, 16436638, 22810475). A similar variant (c.533C>G) resulting in the same protein effect observed here has been observed in individual(s) with clinical features of juvenille polyposis syndrome (PMID: 10441006). This variant is not present in population databases (ExAC no frequency).
