Uncertain significance for DNA ligase IV deficiency — the classification assigned by Clinical Genetics and Genomics, Karolinska University Hospital to NM_206937.2(LIG4):c.94C>T (p.Arg32Cys), citing ACMG Guidelines, 2015: The variation shows a moderately conserved nucleotide (phyloP: 4.97 [-19.0, 10.9]) and is situated in the DNA ligase, ATP-dependent, N-terminal domain. There is a large physicochemical difference between Arg and Cys. The c.94C>T variant is present in population databases (rs770549827, gnomAD v2.1.1 - 5 heterozygous allele counts and gnomAD 4.0.0 - 43 heterozygous allele counts ). This variant has t oour knowledge not been reported in the literature in individuals affected with LIG4-related conditions. Computational predictions do not agree on the potential impact of this missense change. In summary the variant is classified as a variant of uncertain significance.
