NM_001127222.2(CACNA1A):c.3479C>G (p.Ala1160Gly) was classified as Uncertain significance for Developmental and epileptic encephalopathy, 42; Episodic ataxia type 2 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with CACNA1A-related conditions. This sequence change replaces alanine with glycine at codon 1161 of the CACNA1A protein (p.Ala1161Gly). The alanine residue is moderately conserved and there is a small physicochemical difference between alanine and glycine.
