NM_006231.4(POLE):c.3493G>T (p.Asp1165Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1165Y variant (also known as c.3493G>T), located in coding exon 29 of the POLE gene, results from a G to T substitution at nucleotide position 3493. The aspartic acid at codon 1165 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
