Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.1060A>C (p.Lys354Gln), citing Ambry Variant Classification Scheme 2023: The p.K354Q variant (also known as c.1060A>C), located in coding exon 7 of the DICER1 gene, results from an A to C substitution at nucleotide position 1060. The lysine at codon 354 is replaced by glutamine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
