NM_024426.6(WT1):c.1015A>G (p.Asn339Asp) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N334D variant (also known as c.1000A>G), located in coding exon 5 of the WT1 gene, results from an A to G substitution at nucleotide position 1000. The asparagine at codon 334 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
