Uncertain significance for Developmental and epileptic encephalopathy, 12 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015192.4(PLCB1):c.319T>C (p.Tyr107His), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PLCB1 gene (transcript NM_015192.4) at coding-DNA position 319, where T is replaced by C; at the protein level this means replaces tyrosine at residue 107 with histidine — a missense variant. Submitter rationale: This sequence change replaces tyrosine with histidine at codon 107 of the PLCB1 protein (p.Tyr107His). The tyrosine residue is moderately conserved and there is a moderate physicochemical difference between tyrosine and histidine. This variant is present in population databases (rs759519379, ExAC 0.004%). This variant has not been reported in the literature in individuals affected with PLCB1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
