Uncertain significance for Lysinuric protein intolerance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003982.4(SLC7A7):c.1507_1517del (p.Pro503fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Pro503Glyfs*5) in the SLC7A7 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 9 amino acid(s) of the SLC7A7 protein. This variant is present in population databases (rs777950149, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with SLC7A7-related conditions. ClinVar contains an entry for this variant (Variation ID: 834451). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
